High Performance Liquid Chromatography
(H P L C)
This is one of the most widely used
chromatographic method for herbal drugs standardisation, quality control
analysis, purification of compounds and even structural confirmations. The
various modes of chromatography available for HPLC systems,
applications and recent advances in HPLC methods-based method development
are covered in our lecture series.

We keep adding new equipment to our established stock of
state-of- the-art facilities. The
picture
on the left is a new addition this year that is solely dedicated for
routine standardisation of natural products. We are particularly keen in
standardisations based on known bioactive compounds or markers. Once a suitable
method for the limit of detection, limit of quantification, linearity range,
reproducibility, etc. are established for the
known bioactive compound(s) of crude drugs (e.g. herbal drugs), a
concentration-response profile is constructed from which the presence and/or
amount of active ingredient(s) in drugs can be accurately measured.

The figure above shows the HPLC profile of a known drug-standard
tested at different concentrations. The peak area of each determination is then
plotted against the response (e.g. UV absorption). A straight line equation with
statistical significance must then be plotted from which the concentration of
the bioactive compound(s) in the test sample is established.
We use a minimum of SIX determinations to ascertain
accuracy/precision.
